SNARE Complex Competition at the Neuromuscular Junction
SNAP-8 (acetyl octapeptide-3, CAS: 868844-74-0) is a synthetic SNAP-25 N-terminal analog. SNAP-25 is a component of the SNARE complex — the molecular machinery that enables acetylcholine vesicle fusion at the neuromuscular junction. SNAP-8 competes with endogenous SNAP-25 for SNARE complex binding sites, partially inhibiting ACh release efficiency at the NMJ. Lower ACh release reduces the force of facial expression muscle contractions — attenuating the repeated mechanical stress on overlying skin that deepens dynamic wrinkles.
KLOW Protocol — Expression Wrinkle Research
Dynamic wrinkles (crow's feet, forehead lines, glabellar lines) deepen through repeated expression muscle contraction. SNAP-8 research models partial NMJ inhibition to attenuate this mechanism. Preclinical models showed up to 63% wrinkle depth reduction.
Different Mechanism from GHK-CU
GHK-CU addresses structural skin biology — collagen matrix, fibroblast activity, angiogenesis. SNAP-8 addresses neuromuscular dynamics — ACh at the NMJ. Completely orthogonal pathways. Running both (GLOW + KLOW) covers both structural and dynamic dimensions of skin quality.
Partial NMJ Inhibition
Unlike botulinum toxin (complete NMJ blockade), SNAP-8 models partial, competitive inhibition. SNARE complex function is preserved but with reduced ACh release amplitude — maintaining expression while reducing the mechanical force applied to skin.

Research FAQ
What is the KLOW peptide?
SNAP-8 (acetyl octapeptide-3) — Clavicular's name for the neuromuscular skin layer. It's a SNAP-25 analog that partially inhibits ACh release at the NMJ, reducing expression muscle contraction force and the deepening of dynamic wrinkles.
Can SNAP-8 be used topically?
SNAP-8 has been studied both as an injectable and as a topical preparation (0.1-2% in carrier). As a research compound, both routes have published data. The injectable form provides more precise dosing control.
